Former villain Gru is now an Anti-Villain League (AVL) agent. He and his partner and wife Lucy Wilde are sent to foil the plans of Balthazar Bratt, a former 80s child actor who went evil after his TV show was canceled due to guilty puberty.

Gru manages to stop Bratt from stealing the Dumont diamond, a giant pink diamond, but is unable to capture it. Due to the constant inability to capture Bratt, Gru and Lucy are fired from the AVL by his irrational new director Valerie Da Vinci, following the retirement of former AVL director Silas Ramsbottom. When Gru and Lucy return home, Gru and Lucy reluctantly tell their children, Margo, Edith and Agnes, about their firing, but they assure them that they will find new jobs. Most of Gru's Minions abandon him when he refuses to turn evil despite losing his job and the fact that his old assistant, Dr. Nefario, has accidentally frozen in carbonite. Only the two Minions named Dave and Jerry remain because they are not there when the others leave with their new leader, Mel. Meanwhile, Lucy struggles to adjust to her new role as the mother of 3 girls.

The next morning, when a butler named Fritz arrives home, Gru finds out that he has a twin brother named Dru, who lives in a distant country named Freedonia, who longs to meet him. Shocked to learn that he has a twin, Gru confronts his mother, who reveals that when she and her husband divorced, they vowed never to see each other again and each took a child to raise on their own, shortly after birth. .

The family decides to embark on a journey to meet Dru, and they are with great surprise to discover Dru's great wealth and the enormous size of his house, which Fritz attributes to their pig farming business.

Meanwhile, the Minions are arrested for hacking a talent show set, shortly after they finish performing. Bratt manages to steal the diamond again, intending to use it to power a giant robot and destroy Hollywood. Dru reveals to Gru that their late father was a legendary supervillain known as "the bald terror," whose evil activities are the true source of the family's wealth. While Dru was fired by his father as a shame, he asks his brother to teach him how to be a villain.

When Gru refuses to go back to his old ways, Dru takes him on a ride around Freedonia in his father's technologically advanced car. Meanwhile, Lucy tries to balance her new duties as a mom, which she exposes after turning away a boy named Niko who fell in love with Margo in the middle of a traditional cheese party, while Edith and Agnes stumble upon a local tavern and they learn from the bartender the story of a mythical unicorn hiding in the nearby forest. Gru tries to convince Agnes that unicorns are imaginary, but to avoid hurting her feelings she doesn't contradict her. The next day, Agnes and Edith set off into the woods to follow the mythical "unicorn", only to find a goat with a horn that Agnes adopts and calls "Lucky". Meanwhile, the Minions' nostalgia for Gru causes them to escape from prison and return to their former master.

Gru convinces Dru to steal the diamond, with Gru secretly intending to take it to the AVL, hoping to get Da Vinci to reinstate him and Lucy as agents of the Anti-Villain League. Despite several setbacks, they manage to recover the diamond and are saved by Lucy after narrowly escaping. After discovering Gru's true motives, Dru confronts his brother about his lie. In return, Gru insults him and decides to leave Freedonia, ending their relationship. Bratt, disguised as Lucy, kidnaps the girls and acquires the diamond again. Gru and Dru make their peace and chase after him after finding the real Lucy.
